The Braun lab is a world leading lab in plant network analysis. The adaptive T helper cell compartment is highly specialized in the elimination of specific microorganisms by a broad range of effector functions. A division of labor is implemented on the cellular level by distinct T helper cell subsets, which are defined by their respective cytokine profiles, transcriptional regulation and migration properties. Th17 cells have previously been identified as major players in pathogenic tissue distruction and autoimmunity. We could identify the existence of an anti-inflammatory Th17 cell counterpart in humans that can be distinguished from pro-inflammatory Th17 cells by its ability to pro-duce IL-10, its IL-1β independent priming requirements and distinct TCR repertoire. Modulation of IL-1β signaling following systemic anti-IL-1β treatment in patients with auto-inflammatory syndromes could shift the balance from pro- to anti-inflammatory Th17 cell subsets in vivo and correlated with complete clinical remission. Further studies in our laboratory have now reveaved novel and surprising properties of human Th17 cells. The aim will be to elucidate the molecular, cellular and epigenetic requirements for establishing these novel Th17 cell functionalities and to assess their relevance for human health and disease.
