Development of Chromatographic Strategies for the Purification of Bioactive Lectins from Fungal Mycelium

Lectins are carbohydrate-binding proteins with promising applications in biotechnology, diagnostics, biomedicine, and glycoscience due to their highly specific carbohydrate recognition properties. While numerous lectins have been isolated from fungal fruiting bodies, efficient recovery of intracellular lectins from submerged-cultivated fungal mycelium remains a significant downstream processing challenge. Cell disruption releases a complex mixture of intracellular proteins, structural polysaccharides, nucleic acids, and other cellular components, making selective purification difficult.
This project is part of an ongoing research program focused on developing an integrated downstream process for the recovery and purification of fungal lectins from submerged-grown Agrocybe aegerita mycelium.
The student will investigate and compare different chromatographic approaches for lectin purification and evaluate their suitability as downstream processing steps.
