Student thesis (B.Sc/M.Sc) or internship in the field of genetically encoded reporter development using directed evolution and screening

The Stiel lab (Stiel-lab.de) at the Institute of Biological and Medical Imaging (IBMI) at Helmholtz Munich focuses on enabling this rich variety of tools for optoacoustic (OA) imaging. OA is an emerging methodology bridging light excitation and ultrasound detection towards deep tissue imaging. The physical concept of this technique requires for imaging tools with exclusively tailored photophysics.

Further, we are interested in the detailed functioning of our tools on a molecular level pushing our understanding towards true top-down-engineering approaches.

The group combines methods of screening-based and rational (computational) molecular engineering with biophysical and in vivo validation of the tools as well as structural elucidation of their mechanism. We draw expertise from structural-, molecular-, cell- and chemical biology as well as biophysics.

We are now seeking a highly qualified and motivated bachelor or master student with an interest in protein engineering and technical infrastructure

Your tasks:
In this project you will make libraries of reporter variants and screen them for beneficial variants using high-throughput Optoacoustic screening approaches. The detailed tasks include:
 Molecular cloning and mutagenesis using standard techniques, e.g., Gibson Assembly, error-prone mutagenesis.
 Protein expression in e. coli.
 Screening using a high-throughput screening device. This requires operation of laser sources and technical equipment as well as basic MATLAB operation.
 Protein purification and photophysical characterization with spectroscopy.
 Data evaluation and report in exchange with your supervisors.

Your profile:
 You are currently studying in the Bachelor or Master programs of Biochemistry, Biophysics, Molecular Biology, Microbiology or similar.
 You are a careful and structured worker with previous hands-on experience in life-sciences laboratories.
 You are a team player, resilient and diligent worker and have an eye for details.
 You share our passion for protein engineering.
 You have previous experience working with imaging tools or other various types of laboratory technical equipment (such as lasers, detection devices, data acquisition systems, etc.)
 You have some experience in coding (e.g. Matlab, Python or similar)
 You are proficiency in the English language
 The laboratory work for a thesis or internship should be conducted for a minimum duration of at least 3 months
