Regulation of human T cell responses in the tumor microenvironment by tonicity signals Our group is interested in the cellular immunoregulation of human T cells in health and disease. We particularly focus on questions related to T helper cell differentiation, maintenance of cytokine memories and functional plasticity of T cells in settings of infections as well as chronic inflammatory diseases. We have recently established the existence of pro- and anti-inflammatory human Th17 cell subsets (Zielinski et al. Nature 2012) and have described a novel T helper cell subset, which is specialized in the production of GM-CSF with implications for the pathogenesis of multiple sclerosis (Noster et al., Sci Transl Med 2014). The master’s student will have the opportunity to engage in an exciting novel avenue of immunological research that focuses on modulation of T cell functions by tonicity signals such as salt. Tumors are highly enriched in NaCl for unknown reasons. How this affects the function of T cells in the tumor microenvironment, their cytotoxicity and tumor killing efficiency will be assessed on a phenotypic and mechanistic level. Proteins form the labor force of the cell. Each protein can perform its particular function only if it folds into a specific shape. So-called chaperones intervene, often during protein synthesis, to prevent misfolding. A team led by Professors Johannes Buchner of the Technische Universitaet Muenchen (TUM) and Don C. Lamb of the LMU Munich have now taken a closer look at how a major chaperone called BiP performs this role. The researchers specifically asked how BiP’s conformation changes from the moment it binds to a substrate up until the substrate is released and how the co-chaperone ERdJ3 influences its functional cycle. They present their findings in Nature Structural & Molecular Biology (online ahead of print, January 9, 2011).

The Technische Universität München (TUM) is seeking applications from highly motivated candidates for PhD (Dr. rer. nat.) positions in hyperpolarised magnetic resonance (MR) imaging. Hyperpo¬larised 13C metabolic imaging is a novel technique to investigate cellular processes and physiological parameters minimally invasive. You will be involved in developing and investigating MR methods for hyperpolarised imaging of novel biological contrast mechanisms. The four different positions focus on the following aspects of hyperpolarisation: alternative 13C‐labelled agents, advanced MR meth¬ods for an efficient signal acquisition and reconstruction . Overall goal is to advance the diagnostic information of MR for applications including oncology annd cardiology. Depending on the specific task this will require good physical, biological and chemical understanding. Candidates having previous experience in magnetic resonance will be preferred. You will be collaborating with a team of experienced scientists from the MR group at GE, the Medical Engineering (Prof Haase), Chemistry (Prof Glaser) and Nuclear Medicine (Prof Schwaiger) depart-ments at TUM, get unique access to latest hardware, as well as an extensive internal and external network of collaborating sites. This theory/simulations project is focused on developing a unifying framework to describe protein aggregation phenomena in solution. Proteins are known to self-assemble into a variety of large-scale structures, with different morphologies and different degrees of mechanical stability. However, a molecular-level understanding of what factors/mechanisms drive the self-assembly process and affect the resulting morphologies is lacking. The goal is to link the level of protein-protein interactions (mediated also by the solvent conditions, e.g. pH, co-solutes etc.) with the large-scale structure and morphology of the resulting aggregates. Applications range from neurodegenerative diseases (Alzheimer’s, Parkinson’s) to novel biomaterials (e.g. protein hydrogels). An important focus of the project will be the understanding of mechanisms by which aggregates can be broken down, with evident implications for pharmaceutical and clinical research. The thesis work consists mainly of analytical theory using tools of statistical physics like master kinetic equations, stochastic and reaction-diffusion methods, backed by coarse-grained simulations. The student will work in the Soft Matter Theory group at the Physics Departement. The work will be done in close collaboration with the experimental groups of Prof. V. Fodera’ and Prof. B. Vestergaard at University of Copenhagen, and Dr. T. Knowles at University of Cambridge. Candidates should hold a Master degree in Physics, Chemistry, Theoretical Biology, Chemical Engineering and have strong interests in theory, modeling and both analytical and numerical calculations.
