Master’s / Medical Doctoral Thesis Opportunity 16.03.2026 – Doktorarbeiten für Humanmediziner und Masterarbeiten We are offering an exciting opportunity for a highly motivated student to contribute to an ongoing project investigating the challenges of CAR-T cell therapy in the context of Pancreatic Ductal Adenocarcinoma (PDAC). ________________________________________ Project Overview Pancreatic ductal adenocarcinoma (PDAC) is characterized by profound resistance to therapy. Despite advances in systemic treatments, the five-year survival rate remains below 15%. Cellular immunotherapies such as Chimeric Antigen Receptor T-cell Therapy have shown remarkable success in hematologic malignancies, but their application in solid tumors has been limited by antigen heterogeneity, a dense stromal barrier, and T-cell exhaustion within the immunosuppressive tumor microenvironment. This study aims to address these barriers by developing CAR-T cells that will be evaluated in patient-derived organoid (PDO) co-cultures and in a Chorioallantoic Membrane Assay xenograft model. The research group of AG Schmidts has extensive expertise in CAR-T cell design, largely due to Dr. Andrea Schmidts’ long-term scientific experience. This project is a collaboration between AG Reichert and AG Schmidts. Through Prof. Reichert’s research group, we have access to a curated biobank of PDAC PDOs derived from surgical resections and biopsy specimens. Organoid cultures faithfully recapitulate key histological and molecular characteristics of the original tumors while preserving intratumoral heterogeneity. Using a panel of PDAC organoid cultures, we are able to evaluate the efficacy of our CAR-T cells in a robust experimental system across diverse tumor backgrounds. Co-culture experiments with PDAC PDOs and CAR-T cells provide a biologically relevant platform to investigate CAR-T cell proliferation, cytolytic activity, and exhaustion dynamics. To further assess therapeutic potential in a more complex setting, the project employs an in vivo xenograft approach using the chicken CAM model. This system provides a rapid and accessible platform for evaluating CAR-T cell performance prior to more advanced preclinical studies. Furthermore, the project explores strategies targeting tumor-associated extracellular matrix components. Specifically, oncogenic splice variants overexpressed in PDAC stroma are being investigated as tumor-selective CAR-T targets. By combining advanced organoid models, targeted CAR-T engineering, and microenvironment-focused strategies, this research aims to develop combinatorial CAR-T therapies for pancreatic cancer. Ultimately, the goal is to improve CAR-T cell infiltration and cytolytic efficacy and to establish translational approaches that may significantly expand the therapeutic potential of cellular immunotherapy in PDAC and other resistant solid tumors. ________________________________________ Your Responsibilities • Culture tumor cell lines • Maintain patient-derived organoids • CAR-T cell production (Cloning and large-scale plasmid preparation, virus production, T-cell transfection) • Generatex knockout (KO) cell lines using nucleofection • Extract RNA/DNA for conventional PCR and qRT-PCR • Analyze gene expression • Perform flow cytometry experiments on CAR-T cell co-cultures, organoids, and organs isolated from chicken embryos • Perform killing assays (luciferase-, FACS-, or Incucyte-based) ________________________________________ What We Offer • One-month internship before the start of the official contract • An international, collaborative, and dynamic research environment • Direct supervision and mentoring throughout the project • Technical support from a postdoctoral researcher (medical doctor) • Opportunity to contribute to peer-reviewed scientific publications • Participation in journal clubs and project-related meetings Students without extensive wet-lab experience are welcome to apply.
