Location: Rad laboratory, TranslaTUM, Einsteinstraße 25, 81675 München Start: negotiable, as soon as possible Duration: 8-9 months, internship with consecutive master thesis recommended Contact: Sebastian Müller (mueller.sebastian@tum.de), Chiara Thordsen (chiara.thordsen@tum.de) Project description: The bad prognosis of pancreatic cancer patients is linked to the early and frequent metastasis of the disease. Despite its clinical importance, the genetic basis underlying pancreatic cancer metastasis is however not well understood. Over 90% of pancreatic cancers harbor an oncogenic KRAS mutation which is the initial driver of pancreatic carcinogenesis. In previous work of our group, we identified that amplification of oncogenic KRAS copy number during early tumor evolution is an important driver of metastasis formation (Müller, et al., Nature, 2018.). Based on these observations and other analyses run at the lab, we now aim to comprehensively characterize a large panel of mouse pancreatic cancer cell lines in various functional in vitro assays. We selected each assay to reflect a specific step of the metastatic cascade. These assays have been already established during the last year and we now aim to systematically link the behavior of cell lines in each assay to their molecular and phenotypic characteristics for improving the understanding of pancreatic cancer metastasis. Qualifications: We are looking for a highly motivated student with a background in cellular/molecular biology or related fields to support our efforts in advancing this project. Since the project is largely based on learning and conducting various cell culture assays, we require a solid foundation in cell culture techniques. If this is not the case, please do not apply. Communicate skills in English will be necessary for working in an international research team. Desired qualifications (but not necessary): microscopy experience and cancer biology background. What we offer: We offer: an excellent scientific supervision (postdoc + PhD student) and laboratory infrastructure with the lab being located at the translational cancer research center (TranslaTUM). You will learn various in vitro metastasis assays and be exposed to CRISPR/Cas9 and Next-Generation-Sequencing techniques that we routinely use in our team. Furthermore, you will have the chance to work in an interdisciplinary and multicultural research team.
